单位定义
1单位定义为在50μl1X Taq DNA连接酶缓冲液中10分钟后加入1μg用Sma I和Sal I切割的Lambda DNA的12个碱基粘性末端的50%所需的Taq DNA连接酶的量在45℃孵育。
质量控制分析
单位表征测定
单位活性使用2倍系列稀释法测量。在1X Taq DNA连接酶反应缓冲液中进行酶批次的稀释并加入到50μL含有λHind III消化的DNA和1X Taq DNA连接酶反应缓冲液的反应中。将反应在45℃孵育10分钟,停止,并在用溴化乙锭染色的0.8%琼脂糖凝胶上分析。
SDS-Page(物理纯度评估)
将2.0μL浓缩的酶溶液加载到变性的4-20%Tris-甘氨酸SDS-PAGE凝胶上,该凝胶侧翼为宽范围的MW标记和2.0μL的1:100稀释的相同酶物种。电泳后,将凝胶染色并比较样品以确定物理纯度。该测试的验收标准要求浓缩样品中污染带的总量不超过稀释样品中目的蛋白质条带的质量,确认浓缩样品的纯度大于99%。
污染测试
单链核酸外切酶活性
将含有10,000cpm放射性标记的单链DNA底物的50μL反应液和10μL在37℃下孵育4小时的酶溶液导致TCA可溶性计数的释放小于5.0%。
双链核酸外切酶活性将含有5,000cpm放射性标记的双链DNA底物和10μL酶溶液的50μl反应液在37℃下温育4小时,导致TCA可溶性计数的释放少于1.0%。
双链核酸内切酶活性将含有0.5μgpENZuC DNA和10μL酶溶液的50μL反应物在37℃下温育4小时,结果通过琼脂糖凝胶电泳测定没有肉眼可察觉地转变为带切口的环状DNA。
大肠杆菌 16S rDNA污染测试
重复5μL酶溶液样品变性,并在TaqMan qPCR测定中筛选污染性大肠杆菌基因组DNA,使用对应于16S rRNA基因座的寡核苷酸引物。测试的验收标准是由3个重复的无模板对照样品的平均值产生的阈值循环计数(C t)。基于无模板对照 Ct值和标准曲线数据之间的相关性,该测定的检测限为<10拷贝基因组/样品。
