Our Viral Direct Buffer A and B are formulated to release viral DNA or RNA from viral particles and maintain the integrity of released DNA or RNA without a degradation issue. In addition, our formulated buffers allow direct PCR or RT-PCR without a need of nucleic acid purification. Our solution-based application for direct viral testing has many advantages over SiO2 filter or magnetic bead based nucleic acid purification: fast, low cost, no loss of DNA or RNA and low variability during solid/liquid phase transition from traditional purification.

DNA提取EZ-Kit是一种简化的NaI,可从组织提取物,血液样品,细胞裂解物,蛋白质或生物制药样品中纯化出几微克的DNA。整个过程包括在一个微量离心管中进行两个简单的孵育和离心步骤,而不使用任何有害物质,如苯酚和CHCl3。优化的洗涤剂可在高蛋白浓度下提取DNA,或在必要时增强蛋白酶K消化。纯化的DNA足够纯化许多DNA应用,例如PCR,实时PCR,限制性内切酶消化和克隆,DNA测序或Picogreen荧光染色。

 

试剂盒组成:

100uL糖原溶液

1ml洗涤剂组合液

25ml NaI溶液

30 mL洗涤缓冲液(无酒精)

 

试剂盒不包含:

乙醇(洗涤缓冲液)

异丙醇(用于DNA沉淀)

2ml微复合管(离心或等效的)

微型离心机(离心或等效的)

 

溶液的制备:

q 加70ml乙醇洗涤液和混合

q 加50uL糖原NaI溶液

q 加2uL糖原洗涤液

 

DNA提取程序:

1、每个样品添加500uL或稀释至2 mL微离心管

q 2、加20uL洗涤剂组合液到每管,并轻轻的涡旋

q 3、选项:如果高浓度的蛋白质干扰DNA提取和DNA分析,每个样品中吸取添加20uL消化蛋白酶K(10mg/ml),在60°C, 20min,然后在95°C灭酶 5min

4、加500uL NaI溶液到每个微管中,并轻轻的涡旋

q  5、50°C孵育10分钟

q 6、加900uL异丙醇涡旋

q 7、室温孵化30min

8、离心机12000转15min

9、轻轻倒出或吸出上清液

q 10、添加1.8ml洗涤缓冲液(含酒精),涡旋

11、离心机12000转10min

q  12、轻轻倒出或吸出上清液

q 13、风干DNA

q 14、添加20-60uL水轻轻涡旋溶解DNA

q 15、纯化的DNA可进行PCR、实时定量PCR、酶切、DNA测序、荧光染色法
