The fluorescent control liposomes contain the lipophilic dye DiA incorporated in the bilayers. It has comparable physical and chemical attributes as for the Clophosome® except no active is encapsulated inside. It is used for tracking the cellular uptake of the liposomes by common methods like confocal, FACS, etc.
The lipophilic aminostyryl probe DiA inserts into liposome bilayers with its two alkyl tails and its fluorophore oriented parallel to the phospholipid acyl chain. When this dialkylaminostyryl probe binds to membranes, it exhibits a strong fluorescence enhancement; its fluorescence in water is minimal.

The fluorescent control liposomes contain the lipophilic dye DiD incorporated in the bilayers. It is used for tracking the cellular uptake of the liposomes by common methods like confocal, FACS, etc.
The far-red fluorescent, lipophilic carbocyanine DiD is a longer-wavelength DiI analog. It is an oil at room temperature and weakly fluorescent in water but highly fluorescent and quite photostable when incorporated into membranes. It has an extremely high extinction coefficient and short excited-state lifetimes (~1 nanosecond) in lipid environments.

The fluorescent control liposomes contain the near IR fluorescent dye, lipophilic carbocyanine DiOC18(7) (“DiR”) incorporated in the bilayers. It has comparable physical and chemical attributes as for the Clophosome® -A except no clodronate encapsulated inside. It is used for tracking the cellular uptake of the liposomes by common methods like confocal, FACS, NIR imaging etc.
The near IR fluorescent, lipophilic carbocyanine DiOC18(7) (“DiR”) is weakly fluorescent in water but highly fluorescent and quite photostable when incorporated into membranes. The sulfonate groups incorporated into this DiI analog improves water solubility. It has an extremely high extinction coefficient and short excited-state lifetimes (~1 nanosecond) in lipid environments.

作用机制：
Clophosome®-A（氯弗松），带负电荷的氯膦酸脂质体，是*代中性氯氟松Clophosome®-N的升级产品，属于第二代的体内巨噬细胞清除剂（in vivo macrophage depletion），本质就是包裹氯膦酸分子的大单室脂质体（LUV）。结合游离氯膦酸不易穿透脂质体的磷脂双分子层和细胞膜，但脂质体可轻易被巨噬细胞吞噬的特点。氯氟松Clophosome被吞噬后，在溶酶体磷脂酶作用下，脂质体的磷脂双分子层被破坏，使得其内的水溶性药物–氯膦酸释放进入细胞。当氯膦酸在细胞内累积，且达到刚发挥药效的阈浓度（threshold concentration），细胞不可逆损伤，通过凋亡诱导进而死亡。而死后的巨噬细胞释放的游离氯膦酸，循环系统中半衰期极其短暂，泌尿系统循环中快速清除【见图1. 巨噬细胞自sha途径】。
如果脂质体能够充分注射进入体内，机体不同来源的巨噬细胞可被清除，包括肝脏（Kupffer cells，库普弗细胞），脾脏（不同的巨噬细胞亚群），肺脏（肺泡但不是小肠巨噬细胞），腹膜腔，淋巴结，关节（吞噬滑膜衬里细胞）和睾wan。
图1：巨噬细胞自sha流程。包覆氯膦酸分子【方形标识】的脂质体通过内吞作用被巨噬细胞消化。之后与溶酶体（L标识）融合，其内的磷脂酶（箭头标识）破坏脂质体的双分子层。越多双分子层被破坏，释放进入细胞的氯膦酸浓度更高。细胞之后通过氯膦酸介导的凋亡作用而杀死。（N=巨噬细胞细胞核）

FormuMax提供两种预埋有氯磷酸盐分子的中性脂质体产品，该产品单次静脉或腹腔注射后可在脾脏经巨噬细胞清除率可达80-90%，并具有高度活性、稳定的理化性质（长期冰箱存放后仍保持均一性及悬浮特性），粘滞度低使用简便，适用于静脉、腹腔、皮下、鼻腔、气管等多种方式给药。

Clophosome®– Clodronate Liposomes (Neutral)
Clophosome®, neutral clodronate liposomes, offers highly efficient in vivo macrophage depletion. Clodronate molecules are encapsulated in small MLV liposomes for superior activity, physical and chemical stability, and convenience of use. It is formulated to an exceptionally high clodronate/lipid ratio. Free clodronate is cleaned out and the liposomal clodronate encapsulation efficiency is more than 90%. Clophosome liposomes remain homogenous and suspended even after long term storage in fridge. It is not viscous and easy to inject. It is manufactured sterile for long term shelf-life.
The product is suitable for dosing with various routes including intravenous, intraperitoneal, subcutaneous, intranasal, intratracheal and etc. We have achieved 80-90% macrophage depletion in spleen (red pulp macrophages) 24 hours after a single 0.2 mL intravenous injection. A starting dose of 0.15 – 0.2 mL (i.v. or i.p.) for 20-25g animal body weight is recommended for good consistency in macrophage depletion. Optimization is recommended for your specific applications.

We are very excited to announce the introduction of our brand new product, Trans-HiTM, as a superior transfection reagent to compete against the leading products in the market. Trans-HiTM is created based on a proprietary polymer technology. It forms stable complex particles with DNA and rapidly releases the DNA molecules upon endocytosis into the cytoplasm. It offers superior and reproducible transfection efficiency on the common cells such as Hela, CHO, HEK293 and many hard-to-transfect mammalian cells with minimal toxicity. Trans-HiTM is very cost-efficient and 1.0mL is sufficient for over 650 transfections in 24 well plates.

ClophosomeTM-氯膦酸二钠脂质体（中性）
ClophosomeTM，氯膦酸二钠脂质体，能地耗竭体内巨噬细胞。氯膦酸二钠分子被封装于小型多层脂质体中（MLV），具有高活性、理化稳定性、使用方便的特点。该剂型具有十分高的氯膦酸二钠/脂质比率。游离的氯膦酸二钠被清除，使得脂质体包封率超过90%。即使在冰箱中长期贮存后，该脂质体仍能保持均一悬浮状态。该品不粘稠，易于注射，且经过无菌过滤，保质期长。适用于多种给药途径，包括静脉注射、腹腔注射、皮下注射、鼻内给药、气管给药等。在单次静脉注射0.1mL24小时后，脾脏内巨噬细胞（红髓巨噬细胞）消除率为80-90%。为保证巨噬细胞耗竭的良好的一致性，对于体重为20-25g的动物，建议起始剂量为0.1-0.15mL（iv或ip）。我们建议您根据您的具体用途加以优化。
