说明

ON-TARGET 非靶向对照池是人,小鼠和大鼠细胞中RNAi实验的理想阴性对照。四组分ON-TARGET 非靶向siRNA被设计为利用种子区域优化和ON-TARGET 修饰模式具有比传统设计的阴性对照siRNA更少的脱靶,并通过基因组微阵列分析进行筛选。用这些对照处理的细胞中mRNA或蛋白质水平的变化反映了可以与用靶特异性siRNA处理的细胞中的水平进行比较的基线细胞反应。

通过可靠的基因沉默和修饰技术减少脱靶效应;提供针对人类、小鼠或大鼠的预设计 ON-TARGETplusSMARTpool 和 siRNA 试剂。

CHEK2 检查点激酶2

为了应对DNA损伤和复制阻滞,通过控制关键细胞周期调节剂停止细胞周期进程。由这个基因编码的蛋白质是细胞周期检查点调节因子和推定的肿瘤抑制因子。它含有与DNA损伤相应的激活必需的叉头相关的蛋白质相互作用区域,并且响应于复制区域和DNA损伤而迅速磷酸化。当被激活时,已知编码的蛋白质抑制CDC25C磷酸酶,防止进入有丝分裂,并且已经显示稳定肿瘤抑制蛋白p53,导致G1中的细胞周期停滞。此外,这个蛋白与BRCA1相互作用并磷酸化,允许BRCA1在DNA损伤后恢复存活。这个基因的突变已经与Li-Fraumeni综合征相关,Li-Fraumeni综合征是通常与TP53中遗传突变相关的高度渗透性家族性癌症表型。此外,这个基因的突变被认为赋予肉瘤,乳腺癌和脑肿瘤的易感性。这种核蛋白是丝氨酸/苏氨酸蛋白激酶的CDS1亚家族的成员。已经发现了编码不同同种型的几种转录本变体。

单独的位置不是CRISPR靶位点功能的指示

沿着VCP基因的编码序列的长度评估crRNA的功能性表明没有特定的模式,因为它涉及外显子位置,增强了设计算法表征功能特征的重要性。重组U2OS Ubi [G76V] -EGFP -Cas9细胞用266种不同的合成crRNA:tracrRNA复合物转染,靶向VCP基因沿着基因编码区的长度(使用DharmaFECT 4 Transfection Reagent,0.07μg/孔和50 nM合成crRNA) :tracrRNA终浓度)。72小时后,使用Envision读板仪(Perkin Elmer)测量EGFP荧光。
